Temozolomide is a medicinal drug, which is a triazene analog of dacarbazine with antineoplastic activity. It acts as a cytotoxic alkylating agent, which is converted at physiologic pH to the short-lived active compound, monomethyl triazine imidazole carboxamide. This cytotoxicity of MTIC is primarily to methylation of DNA at the O6 and N7 positions of guanine results in the inhibition of DNA replication. It is an active component is metabolized to MITC at all sites.
