Active Ingredient: The main active ingredient in this medicine is Entacapone. It is a nitrocatechol with the anti-parkinsonian property. It belongs to a class of drugs known as a selective and reversible inhibitor of catechol-O-methyltransferase. It catalyzes the transfer of the methyl group to the phenolic group of substrates that contain a catechol structure. It helps to prevent the metabolism and inactivation of adjunct drugs so as to increase the bioavailability of the compound by facilitating passage across the blood-brain barrier.
Indications: This medicine is indicated with other medications such as levodopa, carbidopa for the treatment of Parkinson’s disease. Comtan is composed of COMT inhibitors. The active component in this medicine blocks a certain natural substance i.e. COMT enzyme that breaks down the levodopa in the body. This particular effect allows the levodopa to last longer in the system so that it doesn’t wear off before the next dose.
Action Mechanism: The active ingredient in this medicine is believed to be through its ability to inhibit COMT and alter the plasma pharmacokinetics of levodopa. When Comtan is given in conjunction with levodopa and an aromatic amino acid decarboxylase inhibitor. It is believed that a number of times levodopa administration, sustained plasma levels of levodopa leads to more constant dopaminergic stimulation in the brain. It results in greater effects on the signs and symptoms of Parkinson’s disease.
