Olanzapine is a thienobenzodiazepine that shows antipsychotic, antinausea, and antiemetic activities. This medication is a selective monoaminergic antagonist that binds with high-affinity binding to the receptors such as serotoninergic, dopaminergic, muscarinic M1-5, histamine H1, and alpha-1-adrenergic. These receptors bind weakly to gamma-aminobutyric acid type A and beta-adrenergic receptors.
This agent appears to the blockade of 5-HT2 and 5-HT3 receptors for serotonin antipsychotic activity is through antagonism to dopamine D2 receptors with rapid ligand-receptor dissociation kinetics. It helps to minimize extrapyramidal symptoms.
